چکیده
212 CYCLOPHOSPHAMIDE-INDUCED CHRONIC CYSTITIS ENHANCES PAIN BEHAVIOR AND BLADDER CAPACITY REDUCTION ELICITED BY RESINIFERATOXIN IN RATS Tomohiko Oguchi, Osamu Nishizawa, Naoki Yoshimura The authors note that while rats with chronic cystitis induced by repeated cyclophosphamide (CYP) injections have been used for basic research of hypersensitive bladder disorders including BPS/IC, changes in pain behaviour and voiding function in this model have not been well characterized. In this study, they therefore evaluated voiding and nociceptive behaviour following intravesical application of resiniferatoxin (RTX) to see whether frequent voiding and pain sensation are increased in these rats. They found that while bladder overactivity usually seen in rats with acute cystitis subsided after repeated CYP treatments, rats with CYP-induced chronic cystitis exhibit bladder hypersensitivity resulting in increased pain behaviour and smaller bladder capacity when afferent pathways are stimulated by RTX (TRPV1 stimulator). They consequently concluded that this chronic cystitis animal model can be considered to be suitable for studying pathophysiological mechanisms inducing chronic pelvic pain conditions in hypersensitive bladder disorders including BPS/IC.

Effect of Lycopene on Cyclophosphamide-Induced Hemorrhagic Cystitis in Rats
Background: Cylophosphamide is used alone or in combination with other drugs for treatment of neoplastic diseases. Hemorrhagic cystitis is a major potential toxicity and dose limiting side effect of cyclophosphamide. The aim of this study was to evaluate the effects of lycopene compared with some antioxidants for the prevention of cyclophosphamide induced hemorrhagic cystitis in rats. Methods: ...
متن کاملRole of CXCR2 and TRPV1 in functional, inflammatory and behavioural changes in the rat model of cyclophosphamide-induced haemorrhagic cystitis.
BACKGROUND AND PURPOSE Cyclophosphamide induces urotoxicity characterized by the development of cystitis, which involves bladder overactivity and inflammation. Here, we investigated the roles of chemokine receptor 2 (CXCR2) and transient receptor potential vanilloid 1 (TRPV1) channels in a rat model of cyclophosphamide-induced cystitis. EXPERIMENTAL APPROACH Cystitis induced by cyclophosphami...
متن کاملA rat long‐lasting cystitis model induced by intravesical injection of hydrogen peroxide
Novel longer lasting inflammatory bladder animal models are needed to better understand the pathophysiology of chronic cystitis. We previously developed a relatively long-lasting mouse cystitis model by intravesical injection of hydrogen peroxide (H2O2). To further evaluate its pathophysiology, in this study, we established and analyzed a rat cystitis model. Under anesthesia, 1.5% H2O2 solution...
متن کاملHerpes Simplex Virus Vector-Mediated Gene Delivery of Poreless TRPV1 Channels Reduces Bladder Overactivity and Nociception in Rats.
Increased afferent excitability has been proposed as an important pathophysiology of interstitial cystitis/bladder pain syndrome (IC/BPS) and overactive bladder (OAB). In this study, we investigated whether herpes simplex virus (HSV) vectors encoding poreless TRPV1, in which the segment in C terminus of TRPV1 receptor is deleted, suppress bladder overactivity and pain behavior using a rat model...
متن کاملBlocking mammalian target of rapamycin alleviates bladder hyperactivity and pain in rats with cystitis
BACKGROUND Bladder disorders associated with interstitial cystitis are frequently characterized by increased contractility and pain. The purposes of this study were to examine (1) the effects of blocking mammalian target of rapamycin (mTOR) on the exaggerated bladder activity and pain evoked by cystitis and (2) the underlying mechanisms responsible for the role of mTOR in regulating cystic sens...
